Title: Comparison of three methods for preparing the biofilm produced by Klebsiella pneumoniae
Abstract: Objective To compare three methods for preparing the biofilm of Klebsiella pneumoniae in vitro. Methods K. pneumoniae biofilm was prepared through high molecular membrane agar method, high molecular membrane broth method and infusion tube broth method. The number of colonies in biofilm was counted to judge the utility of each method. Results The colony count of above three methods were (1.32±0.26)×10~ 9 CFU/mL, (0.22±0.026)×10~ 9 CFU/mL and (0.15±0.025)×10~ 9 CFU/mL respectively after 5 days incubation and (3.68±0.51)×10~ 9 CFU/mL, (0.24±0.022)×10~ 9 CFU/mL and (0.20±0.035)×10~ 9 CFU/mL after 10 days. High molecular membrane agar method was much better than the other two methods (P0.01). Conclusions High molecular membrane agar method is a convenient and effective method to prepare K. pneumoniae biofilm.
