Abstract:A method for the HPLC determination of soybean isoflavones in health-care foodstuffs is proposed.With the aid of an orthogonal test,the optimum acid hydrolysis conditions were obtained as follows:HCl ...A method for the HPLC determination of soybean isoflavones in health-care foodstuffs is proposed.With the aid of an orthogonal test,the optimum acid hydrolysis conditions were obtained as follows:HCl conˉcentration in80%(φ)methanol,2.0mol/L;acid volume,40mL;hydrolysis temperature,80℃;hydrolysis duration,3h.Hydrolyzed samples were then analyzed by HPLC through a Hypersil ODS2C 18 column(250mm×4.6mmID,5μm).The mobile phase employed was methanol-water(47∶53),and its flow rate was taken to be1.0mL/min,while the column temperature was selected at40℃.The detection wavelength was260nm.The linear range was0.5~96mg/L with a correlation coefficient of0.999.The relative standard deviation(RSD)was found to be2.09%,and the average recoveries were98.5%~100.5%.Read More
