Title: Establishment of two types of chronic heart failure models in rats
Abstract: Objective To explore the characteristics of two experimental models of chronic heart failure( CHF) established by coronary artery ligature and abdominal aorta constriction separately in rats. Methods A total of 60 Sprague Dawley rats were randomly divided into four groups: coronary artery ligature group,abdominal aorta constriction group,sham coronary artery ligature group and sham abdominal aorta constriction group. The rats in sham coronary artery ligature group and sham abdominal aorta constriction group had no ligaturing coronary artery or narrowing abdominal aorta and the rest of the operation was the same as the coronary artery ligature group or the abdominal aorta constriction group. Hemodynamics and plasma brain natriuretic peptide( BNP) were detected eight weeks after the operation. Results Compare with corresponding sham-operation group,left ventricular systolic pressure( LVSP) and left ventricular pressure maximum rising and falling rates( ± dp / dtmax)decreased significantly( P 0. 01),while left ventricular end-diastolic pressure( LVEDP) increased significantly in coronary artery ligature group and abdominal aorta constriction group( P 0. 05). Plasma BNP level in coronary artery ligature group and abdominal aorta constriction group was significantly higher than that in corresponding sham-operation groups( P 0. 01).Compared with abdominal aorta constriction group,the LVSP and ± dp / dtmaxin coronary artery ligature group were significantly lower while LVEDP and plasma BNP were significantly higher( P 0. 05). Conclusion Both coronary artery ligature and abdominal aorta constriction are useful methods to establish experimental models of CHF,BNP increased significantly in the both operation groups.
