Title: RP-HPLC Determination of Quercetin in Eupatorium lindleyanum
Abstract: ObjectiveAn RP-HPLC method was established for the determination of quercetin in Eupatorium lindleyanum,to compare the content of quercetin in Eupatorium lindleyanum from different product area.MethodsRP-HPLC was used to determine the content of quercetin in Eupatorium lindleyanum after herb was extracted and hydrolyzed.Quercetin was separated completely on a Komasil C_(18) column(4.6 mm×250 mm,5 μm),using a mixture of methanol-0.4%phosphoric acid(50∶50) as mobile phase.With UV detection at 360nm,and the flow rate is 0.6 ml/min,the temperature of the column is 30℃.ResultsThe calibration curve was linear in the range of 2.78~27.8 μg/ml of quercetin with correlation coefficient being(0.999 13).The average recovery was 98.698%(RSD = 2.4%,n=5).ConclusionThe method is quick and simple with good repeatition and can be used for quality control.
