Title: The Optimization of Fermentation Conditions of Chitinase-producing Fungus Gliocladium virens F051 and its Degradation Effect on Three Pathogens
Abstract: In this paper,systematical studies were made of the effects of varied factors on chitinase-producing fungus Gliocladium virens F051,including carbon source,nitrogen source,cultural temperature,pH,bottled volume and inoculated volume.The results showed that the optimal carbon source and nitrogen source were maltose and yeast powder.By synthesizing the result of orthogonal design,the best fermentation conditions were got as follows: cultural temperature:28℃,pH 7,100 ml medium volume in 300ml triangle flask bulk,1ml inoculating volume,1.5% maltose and 0.2% yeast powder.According to the effect of time on the hyphae growth and chitinase,the chitinase activity would be the highest on the fourth day.On this basis,the effect of chitinase on three pathogens showed that it could degrade the cell-wall of the pathogens.The chitinase could make the pathogen cell-wall become thin or rupture,cell-wall and bioplasm separate,bioplasm become vacuolate and so on.
