Title: Human Embryo Liver L-02 Cells Damage Induced by Fullerene
Abstract: Objective To study the toxicity and mechanism of fullerene(C_(60))on human embryo liver L-02 cells.Methods L-02 cells were exposed to C_(60)suspension of different concentrations(0.00,1.25,2.50,5.00,10.00,20.00 and 40.00μg/ml)for 24 h,then the content of GSH and the activity of LDH,SOD were determined,the viability of cells with/without NAC was also compared.Results Compared to the control group,the viability of cells exposed to 1.25,2.50,5.00,10.00,20.00 and 40.00μg/ml C_(60)suspension decreased in a dose-dependent manner,and the differences were significant(P0.05 or P0.01).In C_(60)-NAC joint exposure groups,the viability of cells increased in degree in groups of 1.25,2.50,5.00 and 10.00μg/ml,and the differences were significant(P0.05 or P0.01).Compared with the control group,cellular LDH activity increased in 1.25,5.00,10.00,20.00 and 40.00μg/ml groups,SOD activity in 5.00,20.0,40.0μg/ml groups and GSH contents in 10.00,20.00,40.00μg/ml groups decreased significantly(P0.05 or P0.01).Conclusion C_(60)can cause L-02 cells damage and oxidative injury should be concerned in the possible mechanism.
