Title: Antioxidant effects of melatonin on cultured rat cardiomyocytes
Abstract: AIM:To investigate the effects of Melatonin (MT) on cardiomyocytes damaged by H 2O 2. METHODS: The rat cardiomyocytes were cultured in DMEM for 4~5 days. Cultured cells were divided into three groups randomly: control group, H 2O 2 group and MT group. The leakage of lactate dehydrogenase (LDH) was assayed by automatic biochemical analyzer. The content of malondialdehyde (MDA) was determined by measuring thiobarbituric acid reactive substances. RESULTS: Compared with those of the control group, both LDH leakage and MDA content increased significantly(from 6.5±1.6 U/ml to 24.8±1.7 U/ml, and from 1.23±0.05 nmol/10 6 cells to 3.65±0.20 nmol/10 6 cells, P 0.01),cell viability decreased significantly(from 96 0%±1.8% to 60 0%±3.2%, P 0.01). In MT group, MT inhibited the increase of LDH leakage and MDA content induced by H 2O 2 (10.2±1.3 U/ml and 1.59±0.09 nmol/10 6 cells respectively) and increased the cell viability(90 0%±2.2%). CONCLUSION: MT can protect the cardiomyocytes against H 2O 2 induced injury with its antioxidative effect.
