Title: Regeneration of Grape ‘Manicure Finger’(Vitis vinifera) from Its Isolated Explants
Abstract: The effects of different explant types,different hormones and their concentration combinations as well as dark culture time on the regeneration of grape 'Manicure Finger'(Vitis vinifera) were studied.The results showed that adventitious buds could regenerate from the isolated leaves,petioles and stem segments;among them the regenerated rate of adventitious buds from the isolated petioles was maximum.TDZ enhanced the regeneration of adventitious buds from petioles better than 6-BA.IBA was better than NAA and 2,4-D in inducing the regeneration of adventitious buds from petioles.Petiole explants displayed the highest regenerated rate after 3 weeks' dark culture.Among all the media prepared,the optimum media for the regeneration of adventitious buds from leaves,petioles and stem segments were MS + 1.0 mg/L TDZ + 0.1 mg/L IBA,MS + 1.0 mg/L TDZ+0.1 mg/L IBA,MS+1.0 mg/L TDZ + 0.2 mg/L IBA,respectively;their regenerated rates were 60.71%,93.10% and 47.37%,respectively;their propagation coefficients were 5.65,5.91 and 5.00 separately.The regenerated adventitious buds were induced to take roots on 3/4 MS+0.35 mg/L IBA medium,then the full plantlets were cultivated for 35~50 d,and their survival rate reached 90% after being trained and transplanted to the greenhouse.
