Title: Studies on callus inducement and plant regeneration of Hippophae rhamnoides L.
Abstract: Shoots and axillary buds were induced \%in vitro\% from stem apexes and stem segments of fine variety of Hippophae rhamnoides L.on MS and 1/2MS basic medium supplemented with different concentration of IAA,IBA,BA and NAA.When the regenerated buds obtained from the explant were inoculated on MS or 1/2MS basic medium containing 0.3-0.5 mg/L BA and 0.05 mg/L NAA,cluster buds were formed and calluses were induced from the leaflet and young stem.A large number of green prominencies were produced on callus surface in subculture process,and developed gradually to adventitious buds.Hundreds of adventitious buds might be produced on the surface of adventitious bud also on the same medium.When the 3 cm height adventitious bud was cut and transferred onto 1/2MS supplemented with IAA or IBA 0.2 mg/L,roots were induced and intact plants were regenerated.
