Title: Study of Expression and Significant of Hepatocyte Growth Factor in Chronic Renal Failure in Rat
Abstract: Objective To investigate the effect of hepatocyte growth factor(HGF) on the expression of matrix metalloproteinase-2(MMP-2) and its tissue inhibitors of metalloproteinase-2(TIMP-2) in renal tissue of rats with experimental chronic renal failure,and study the possible mechanism of antifibrotic effects of HGF.Methods The rats in adenine-induced model group received 250 mg/(kg·d) by gavage with 2.5% adenine and 2% starch solution.The rats in control group only received starch solution by gavage at the same time.Haematoxylin and eosin(HE) and Masson staining were used to study the dynamic histological changes of the kidneys.The expressions of HGF,MMP-2,TIMP-2 were analyzed by immunohistochemistry staining.Results Renal damages were exacerbated in model group in comparison with those of normal control group at each time point.The expression of HGF was positive related to the expression of MMP-2 and negatively correlated with the expression of TIMP-2.Conclusion The mechanisms of antifibrotic effects of HGF may be associated with MMP-2 and TIMP-2 in renal interstitial progress.
