Title: Inducing-apoptosis effect of Bortezomib on prostate cancer PC-3 cells
Abstract: ObjectiveTo explore the effect of Bortezomib on proliferation and apoptosis of prostatic carcinoma PC-3 cells.Methods MTT assay was used to detect the proliferation,while Annexin-V staining was used to investigate apoptosis of PC-3 cells.RT-PCR was used to analyze the mRNA expressions of bcl-2,bax and bim.Results Bortezomib inhibited the proliferation of PC-3 cells in time-and dose-dependent manners.After having treated with Bortezomib for 24 hours,PC-3 cells showed obvious nuclear condensation,chromatin condensation and fragmentation.Annexin-V-positive rate was 18.5%,significantly higher than that of the control group(2.04%).RT-PCR showed that Bortezomib up-regulated the mRNA expression of bax in 24 h,while down-regulated expression of bcl-2 and bim.Conclusion Bortezomib inhibits the proliferation of PC-3 and induces apoptosis by up-regulating bax and down-regulating bcl-2 and bim.
