Title: Expression and clinical significance of Notch1 and HIF-1α proteins in human gastric carcinoma
Abstract: Objective To explore the relations between expression of Notch1 and HIF-1α proteins in human gastric carcinoma and clinical pathological features and prognosis.MethodsThe expression of Notch1 and HIF-1α proteins were detected in 135 cases of gastric carcinoma and 27 cases of normal gastric mucosa tissue by tissue microarray technique and immunohistochemical EnVision method.The chi-square test was used to analyze the difference of the positive expression rates of Notch1 and HIF-1α proteins between the normal gastric mucosa group and the gastric carcinoma group,and its correlations with the clinical pathological features.The correlation between the positive expression of Notch1 and HIF-1α proteins in gastric carcinoma was assessed by the Spearman rank correlation coefficient.The Kaplan-Meier survival analysis was used to evaluate the relationship between the positive expression of Notch1 and HIF-1α proteins and survival time.ResultsThe positive expression rates of Notch1 and HIF-1α proteins in gastric carcinoma tissue were higher than that in normal gastric mucosa(P0.05).The positive expression rates of Notch1 and HIF-1α proteins in the group of Ⅲ-Ⅳ stages(of TNM stage)and the group with lymphonode metastasis were higher than that in the group of Ⅰ-Ⅱ states(of TNM stage)and the group without lymphonode metastasis,respectively(P0.05).Notch1 expression was positively correlated to HIF-1α protein expression in gastric cancer(P0.01).Their expression was also closely correlated with survival time.The mean survival time of the negative expression group was longer than that of the positive expression group(P0.05).ConclusionThe positive expression of Notch1 and HIF-1α proteins in gastric carcinoma is significantly correlated with the invasion and lymphonode metastasis of gastric carcinoma,which may be of great importance for predicting the prognosis of gastric carcinoma.
