Title: HPLC determination of nuezhenide in Fructus Ligustri Lucidi
Abstract: Objective:To develop a quantitative method for the determination of nuezhenide in Fructus Ligustri Lucidi.Method:An HPLC method was applied to separate and quantify nuezhenide. The samples were separated by an Alltima C18 column(4.6 mm×250 mm,5 μm) with acetonitrile(A)-water(B) as mobile phase[0-40 min,15% A→40% A].The flow rate was 1.0 mL·min-1,and the detection wavelength was set at 230 nm.Results:The linear range of nuezhenide was 50-1000 μg·mL-1(r=0.9999),and the average recovery(n=5) was 99.01%(RSD=0.17%).As the results shown, the content of nuezhenide in Fructus Ligustri Lucidi was 0.24%-3.06% from 10 different habitats. Our study suggests that the content of nuezhenide in Fructus Ligustri Lucidi should not be less than 1.7%.Conclusion:The developed method is sensitive, rapid, accurate and specific. And it can be used to control the quality of Fructus Ligustri Lucidi.
