Title: Comparative study on methods for preparation of models of chronic ischemic heart failure
Abstract: Objectives To compare preparation of the model of chronic heart failure after acute myocardial infarction by cryoinjury of anterior wall of left ventricle or ligation of anterior descending branch in rats. Methods 120 SD rats were randomized into cryoinjury (50), ligation (50) and sham-operated groups(20). ECG and heart function examination were performed 4 weeks after operation. Heart tissues were taken for TTC and HE staining and mortality of the rats was observed. Results Four weeks after operation, the mortality of cryoinjury group was 32%,which was lower than 48% in ligation group. Compared with sham-operated group,ECG showed myocardial infarction,obvious decrease in LVEF and shortening rate of short axis of left ventricle and obvious increase in end diastolic diameter of left ventricle in cryoinjury group and ligation group, the differences had statistical significance (P0.01). TTC stained myocardium section of rats in cryoinjury group and ligation group showed 100% infarction area. Hematoxylin-eosin staining showed scar in myocardial infarction area, while myocardial infarction was not found in sham-operated group. Conclusions Heart function and myocardial infarction size of cryoinjury-induced model were equal to those in ligation-induced model. The mortality of cryoinjury model was lower than that of ligation model. The model induced by cryoinjury of left ventricle anterior wall is better than that induced by ligation of anterior descending branch.
