Title: Inducement of tubule-like structure and EphrinB2/EphB4 expression of human umbilical vein endothelial cells by VEGF
Abstract: Objective To observe the in vitro formation of tubule-like structure (TLS) and expression of EphrinB2/EphB4 by human umbilical vein endothelial cells (HUVECs) induced by vascular endothelial growth factor(VEGF). Methods HUVECs were separated by type Ⅰ collagenase and cultured in a three-dimensional culture system for the TLS formation, exposured to VEGF at concentration from 0.1 ng/ml to 50 ng/ml. Immunocytochemistry was used to examine the effects of VEGF on expression of EphrinB2/EphB4 in HUVECs. Results HUVECs expressed EphB4, but did not express ephrinB2. The expression of EphB4 was not dependent on VEGF concentration. TLS formation was significantly induced by VEGF in a dosage-dependent manner. Conclusion VEGF had the ability to induce HUVECs to form the TLS in three-dimensional culture model and had no stimulation effect on expression of EphrinB2 or EphB4.
