Title: In situ study on rat intestinal absorption kinetics of iriflophenone 3-c-β-D-glucoside(MAH 7-2-4-2-2)in extract of mango leaf
Abstract: Objective To study the rat intestinal absorption kinetics of MAH 7-2-4-2-2 in situ. Methods Single-pass perfusion model was used for rats in situ,the MAH 7-2-4-2-2 content was determined by HPLC and the absorption sites,perfusate concentration were investigated in order to reveal their influences on the absorption of MAH 7-2-4-2-2. Results The statistics showed that the ka and the Papp values of the duodenum,jejunum,ileum,had no significant differences(P0.05).The ka values of drug in the small intestine and colonic had significant differences(P0.05);the Papp values of drug in the small intestine and colonic also had significant differences(P0.05).The concentration from 5.15-102.90 mg·L-1 had no distinctive effect on the ka and the Papp. Conclusions MAH 7-2-4-2-2 could be absorbed at all intestine tract of rats and no highest saturated concentration appeared.Passive diffusion mechanism is proposed as the major factor for the absorption of MAH 7-2-4-2-2.
