Title: Study on Clone Establishment From Tender Stems of Cynanchum Wilfordii
Abstract: In order to preserve the wild resources and meet the needs of medical planting,with the method of tissue culture,tender stems of Cynanchum wilfordii were used as material to do the research on callus inducement and differentiation,rooting of adventitious buds,rooting multiplication of tube seedlings,ransplantation of tube seedlings and stable planting and established the clone from tender stems.The results show that the optimum medium for callus induction was MS+ZT 0.2 mg/L+2,4-D2.0 mg/L,MS+AgNO3 0.6 mg/L+NAA 0.1 mg/L+ZT 0.6 mg/L was suitable for callus differentiation,1/2MS+NO.ABT2 0.8 mg/L+IAA 0.3 mg/L was the best rooting medium for adventitious buds,the ideal medium for rooting multiplication of tube seedlings was 1/2MS+NO.ABT2 0.6 mg/L+IAA 0.2 mg/L,the transplanting survival rate of tube seedlings was 91.9% and stable planting survival rate was 98%,the tube seedlings survivaled retained all botany characteristics of Cynanchum wilfordii with exuberant growth and developed root system,harvest root fresh weight increasing 37% the following year.
