Title: Enhancement of Cisplatin-induced Apoptosis of Human Osteosarcoma MG-63 Cells by Celecoxib
Abstract: Objective To explore the enhancement of cisplatin-induced apoptosis of human osteosarcoma MG-63 cells by celecoxib.Methods Divide MG-63 cells into 3 groups and treat with celecoxib,cisplatin and celecoxib + cisplatin respectively.De-termine the cell activity by MTT method and the proportion of apoptotic cells by flow cytometry.Observe the apoptosis of cells by AO / EB staining and calculate the apoptosis rate.Results After treatment with celecoxib alone,the inhibiting rate of MG-63 cells increased with the increasing concentration of drug.However,the inhibiting rate of cells treated with celecoxib + cisplatin was signifi-cantly higher than that with cisplatin alone.FCM analysis proved that the proportion of apoptotic cells treated with celecoxib + cis-platin increased significantly.AO / EB staining showed that the apoptosis rate of cells treated with celecoxib + cisplatin was signifi-cantly higher than that with celecoxib or cisplatin alone.Conclusion Both celecoxib and cisplatin induced the apoptosis of human osteosarcoma MG-63 cells.However,the combination of the two drugs enhanced the apoptosis significantly.
