Title: Celecoxib on Human Hepatoma Cell Proliferation Inhibition and Radiosensitization
Abstract: Objective To investigate the function of celecoxib on human hepatocellular carcinoma cell huh-7 proliferation inhibition and radiotherapy sensitization.Methods In human liver carcinoma cell line huh-7 as the research object,using celecoxib and high-energy ray as a means of intervention,the function of celecoxib on cell proliferation inhibition was observed in different concentration and action time by MTT,and the apoptosis and cell cycle effect of celecoxib combined with radiotherapy on hepatoma cells were evaluated by using flow cytometric technique.Results ComPared with control group,the absorption value decreased significantly with the increase of celecoxib concentration and exposure time.Celecoxib on liver cancer radiotherapy sensitization effect is obvious.Compared with the control group,drug group increased the proportion of G0/G1 phase cells,reduced the proportion of S phase cells,G2/M had no obvious meaning;G2/M phase cells increased in the radiotherapy group;the combination group increased the proportion of G0/G1 phase cells,S phase cells reduced,Cell proliferation was inhibited.Conclusion Celecoxib can improve the human hepatoma cell line huh-7 sensitivity to the role of radiotherapy,its role may be through the induction of apoptosis,cell cycle phase distribution changes to achieve.
