Title: The effects of Ghrelin on hyperglycemia-induced apoptosis in cultured cardiomyocyte
Abstract: Objective To investigate the effects of Ghrelin on hyperglycemia-induced apoptosis in cultured rat cardiomyocyte.Methods The cultured cardiomyocytes were randomly divided into Group N (cardiomyocytes exposed to low glucose level,5.5 mmol/L);Group H (cardiomyocytes were cultured with high glocose level,25.0 mmol/L);Group G(cardiomyocytes were cultured in both high glucose level,25.0 mmol/L and Ghrelin).Apoptosis in different groups was determined by TUNEL.The change of caspase-3 activity in cardiomyocytes was detected by caspase-3 activity assay kit.Results Compared with that of group N,the number of apoptosis cardiomyocytes was significantly increased in group H.The caspase-3 activity in group H was higher than that in group N.The number of apoptosis cardiomyocytes was significantly decreased in group G relative to group H,and the caspase-3 activity in group G was markedly lower than that in group H.(P0.05).Conclusions Ghrelin could effectively inhibit hyperglycemia-induced apoptosis in cultured rat cardiomyocyte by decreasing caspase-3 activity.
