Title: Establishment of RAPD Reaction System and Analysis of Genetic Diversity for Bryophytes
Abstract: An optimal protocol of Random Amplified Polymorphism DNA(RAPD) is established for Brachythecium fasciculirameum.Based on it,analyses of genetic diversity are conducted with eleven species of bryophytes.The results show that the optimal protocol is accomplished in 25 μL reaction volumes containing 1 unit Tag DNA polymerase,2.0 mmol/L MgCl2,0.2 mmol/L dNTPs,60 ng template DNA and 10 pmol/L RAPD primer.Six primers are selected from forty ones to generate seventy-seven DNA bands.A dendrogram is established based on Hierarchical cluster analysis by 11.5 SPSS,which is similar to morphological studies.It is clear that the RAPD technology can be used to analyze the genetic diversity of bryophytes.
