Title: Optimization of Tissue Culture Conditions for Asparagus cochinchinensis
Abstract: The young stems of Asparagus cochinchinensis(Lour.)Merr.were used as explants to study the disinfection effect of 1 g/L HgCl2 with different treatment times,and the effect of different hormone combinations on induction rate of callus,adventitious buds and rooting.The results showed that the browning and infection rates of explants were low when sterilized for 7 min by 1 g/L HgCl2.The optimum medium for callus induction was MS+0.5 mg/L 6-BA+1.0 mg/L NAA+0.2 mg/L KT.The optimized medium for adventitious buds induction was MS+1.0 mg/L 6-BA+0.2 mg/L NAA+0.2 mg/L KT.The medium with the highest rooting rate(76%) was 1/2MS+0.5 mg/L NAA+0.5 mg/L IBA.
