Title: Determination of Total Flavone in Diospyros Leaves Via Colorimetry
Abstract: To establish the methods of determination of total flavones in diospyros leaves according to colorful chelate formed by flavone and metal ion and choose the optima conditions on sample preparation.Optima conditions of sample preparation were determined by mating design;spectrophotometry was conducted;KAc-AlCl3 system was reacted with flavones extract from samples;the maximum wavelength was selected;standard curve was made with Rutin-the control article;and methodology was investigated.Sample preparation was under the condition of 70 % ethanol,extraction 75 min,and the ratio of solid to liquid was;the content of total flavones was 1.13 % in diospyros leaves via KAc-AlCl3 system colorimetry and RSD was 1.86 %(n=6);the average recovery rate was 99.23 % and RSD was 0.27 %(n=5).The method of determination of total flavone in diospyros leaves via KAc-AlCl3 system colorimetry was feasible with stable color and simple conduction,which can be used for the determination of the total flavonoids and be suitable for quick quality monitoring of diospyros leaves.
