Title: ISSR analysis on genetic variations among different varieties of Lonicera japonica
Abstract: The genetic variation of different varieties of Lonicera japonica was analyzed by inter-simple sequence repeats(ISSR). Ten ISSR primers with high polymorphic and clear bands were f iltered and utilized for testing the genetic variance of 23 Lonicera japonica varieties. The results demonstrate that 174 DNA fragments were amplif ied, among which 169 DNA bands were polymorphic with the ratio of polymorphism 97.1%, 23 genetic similarity coeffi cients ranged from 0.471 3 to 0.862 1, with an average of 0.629 9; Nei's gene diversity(He) and Shannon's information index were 0.379 3 and 0.556 3 at the species level respectively, which proved that the Lonicera japonica varieties had a wide genetic base and a high genetic diversity; The cluster analysis presented that the 23 varieties were divided into three main groups by using the unweighted pair-group method with arithmetic average(UPGMA) based on ISSR molecular marker data. So, the ISSR markers can be effectively used to evaluate the genetic diversity of honeysuckles, thus providing a theoretical foundation for the identifi cation and classif ication of honeysuckle cultivars, and for the eff icient use of their germplasm resources.
