Title: Role of XAF1 gene expression in apoptosis and proliferation of pancreatic cancer SW1990 cells
Abstract: Objective To investigate the effects of exogenous overexpression of X-linked inhibitor of apoptosis(XIAP)-associated factor 1(XAF1)mediated by adenovirus vector Ad5/F35 on the proliferation and apoptosis of pancreatic cancer cell line SW1990. Methods Recombinant Ad5/F35-XAF1 virus was constructed and packed,and pancreatic cancer cell line SW1990 was infected(Ad5/F35-XAF1 group).Besides,control virus infection group(Ad5/F35-Null group) and blank group were established.The infected cells were identified by RT-PCR and Western Blotting.Cell apoptosis and cell cycle were analysed by flow cytometry,cell apoptosis rate was determined by TUNEL,and cell proliferation was detected by MTT assay. Results The expression of XAF1 mRNA and protein in SW1990 cells was significantly increased in Ad5/F35-XAF1 group.Compared with control group and Ad5/F35-Null group,the cell apoptosis rate was significantly increased,the cell proliferation viability was significantly decreased,and the percentage of cells in G2/M phase was significantly higher in Ad5/F35-XAF1 group(P0.05). Conclusion Cell apoptosis is increased and cell proliferation is inhibited in pancreatic cancer cell line SW1990 infected with recombinant Ad5/F35-XAF1 virus.
