Title: Study on hardwood cutting propagation of Fraxinus chinensis clones
Abstract: The research used the 1-year-old branch cutting of Fraxinus chinensis clones under four different treatment: age of cuttings,media,types of auxin,auxin concentration and treatment time to conduct the L9(34) orthogonal experiment and study the technology on hardwood cutting propagation of F.chinensis clones and the activity dynamic variation of important rooting enzyme: indole acetic acid oxidase(IAAO),polyphenol oxidase(PPO) and peroxidase(POD).The results showed that: in the 9 treatments tested,the 9th treatment(1-year-old cutting from 5~6 year-old male plant,mixed medium of pure sand,pure perlite and vermiculite IAA,100 mg/L,2 h treatment) is the best.The rooting process of F.chinensis cutting propagation can be divided into 3 periods: callus formation,adventitious root induction,dventitious root elongation.There was regular change of the three rooting-related enzymes during rooting period.The activity of the 3 rooting enzymes is clearly distinguished in different treatments and rooting phases,indicating that in the rooting process of F.chinensis cutting,the occurrence and elongation of adventitious roots are closely linked with these 3 rooting enzymes'activity.
