Title: The role and relationship of IL-13 and IL-4 in asthma pathogenesis
Abstract: Objective To explore the role and potential relationship of IL-13 and IL-4 in asthma pathogenesis. Methods The blood samples were taken from 24 asthma children and 24 normal children . ELISA was used to detect the levels of IL-4 and IL-13 in culture supernatants of PBMC. Results (1) The level of IL-13 in culture supernatants in 3 d,5 d,7 d was significantly higher than that of 1 d in control and asthma group,the level of IL-13 in 3 d,5 d,7 d of asthma group was significantly higher than that of control,the level of IL-13 in 3 d was significantly higher than those of 5 d and 7 d in control group,but there was no significant difference in the level of IL-13 in 3 d,5 d,7 d in asthma group. (2) The level of IL-4 in 1 d and 3 d was significantly higher than that of 5 d and 7 d in control and asthma group,the level of IL-4 in 1 d,3 d,5 d and 7 d of asthma group was significantly higher than that of control group. (3) The level of IL-13 was significantly lower than that of murine IgG control group in control and asthma group after treated with IL-4 McAb. (4) The level of IL-12 was significantly higher than that of murine IgG control group in control and asthma group after treated with IL-13 McAb.(5) The level of IL-12 was significantly higher than that of murine IgG control group in asthma group after treated with IL-4 McAb and IL-13 McAb. Conclusion It seems that IL-4 may have a role in earlier phase of asthma and can induce IL-13 production. IL-13 may have a role in pathogenesis of asthma. IL-13 McAb phase could induce T H1 derived cytokine in higher level and may have more effective role in the therapy asthma than that of IL-4 McAb.
