Title: Effect of electroacupuncture on the iron levels in the substantia nigra of 6-OHDA lesioned rat
Abstract: Objective To explore the mechanism of the neuroprotective effect of electroacupuncture in the treatment of Parkinson's disease(PD).Methods The rat model of PD with a unilateral injection of 6-OHDA into SNc was adopted.Fifty five healthy male Wistar rats were divided into five groups randomly,normal group,PD group,normal-electroacupuncture group,electroacupuncture group,sham-electroacupuncture group,control group,siRNA control group,and nine rats per group.Rats in each group except normal group were treated with electroacupuncture for two weeks.The effect of electroacupuncture in some special acupoints on the content of iron in the substantia nigra of rat,which was determined by atomic absorption spectrometry(AAS),was investigated.Result Compared with the normal group,the levels of iron in the PD group were significantly higher as well as the net turns of unilateral lesioned rats do.However,compared with the PD group,the levels of iron in the electroacupuncture group were significantly decreased companied with the improvement of rotational behavior.Conclusions Electroacupuncture could decrease the content of iron in the substantia nigra of 6-OHDA unilateral lesioned rat and improve its rotational behavior.The Results in the present study reveal that electroacupuncture may have effect on the regulation in the metabolism of iron in the substantia nigra of 6-OHDA unilateral lesioned rat.It may be one of the mechanisms on the neuroprotective effect of electroacupuncture in the PD treatment.
