Title: Conversion of RAPD marker linked to the flesh color in peach to SCAR marker
Abstract: RAPD technology was used to screen molecular marker linked to flesh color in 69 F1 hybrids derived from the intraspecific cross between peach cultivar Jingyu and Flavortop. The OPU03 primer resulted in a reproducible bright polymorphic band of 899 bp present in the white-fleshed individuals and peach cultivar Jingyu. The polymorphic band was then cloned and sequenced. Two primers were designed based on the sequence to transform into SCAR marker. The RAPD marker was successfully converted into a SCAR marker with primer combination BFP15/60, which was designated SCU03-900. The linkage distance is 21 cM between the SCU03-900 and the flesh color. The SCAR marker will be useful for marker-assisted selection for flesh color in peach breeding programs.
