Title: A study on the effect of celecoxib and cisplatin on the proliferation and apoptosis in ovarian cancer cell line
Abstract: Objective To explore the effect of selective cyclooxygenase-2 inhibitor celecoxib and cisplatin on the proliferation and apoptosis in ovarian cancer cell HO-8910.Methods The proliferation of cells was detected by MTT methods.The distribution of cell cycle and rate of apoptosis were determined by flow cytometry.The expression of Survivin,PCNA and COX-2 protein was detected by immunohistochemistry.Comparisons between groups were performed with one-way ANOVA analysis and LSD-t test.Results Both celecoxible and cisplatin could inhibit the proliferation of HO-8910 cells(P0.01),combined use of celecoxible and cisplatin can enhance the apoptosis rate,increase cells in G0/G1 phase(P0.01),and down-regulate survivin、PCNA and COX-2 expression(P0.01).Conclusion Both celecoxible and cisplatin could induce apoptosis of HO-8910 cells,and their effect is synergetic.
