Title: Ischemic preconditioning inhibit early apoptosis of hepatocytes in reduced-size rat liver transplantation model
Abstract: Objective To investigate the effect of ischemic preconditioning(IPC) on early hepatocellular apoptosis after reperfusion injury in reduced-size liver transplantation model and its significance.Methods We performed 50% sized rat liver transplantation model.A total of 72 adult male SD rats were randomly divided into two groups: control group and IPC group.Serum ALT levels at 2nd,6th and 24th hour were examined and histopathological changes were analyzed.Apoptotic index of hepatocytes in liver grafts at 24 th hour after reperfusion were examined by using TUNEL assay.The expression of bcl-2 and caspase-3 protein were examined by method of immunohistochemistry.Results Compared with control group,we observed significantly changes in IPC group,including serum ALT levels decreased at 6th and 24th hour after reperfusion,minimal liver damaged,apoptotic index of hepatocytes and caspase-3 expression reduced,as well as the expression of antiapoptotic pretein bcl-2 increased.Conclusion IPC could ameliorate early ischemia reperfusion injury in reduced-size liver grafts.It's protective mechanisms might be mediated in part by prohibiting the early hepatocellular apoptosis,which due to the upregulate of bcl-2 and inhibite of caspase-3.
