Title: Characteristic of mutations in rpoB gene of 46 multi-drug resistant Mycobacterium tuberculosis isolates from Fujian Province
Abstract: The aim of this study is to characterize the mutations in rpoB gene of multi-drug resistant Mycobacterium tuberculosis isolates from drug resistant survey in Fujian Province,and analyze the polymorphism of alleles in rpoB gene,providinging the molecular evidence for rapid diagnosis for multi-drug resistant tuberculosis.The isolates were collected from the drug resistant survey sites in Fujian Province,and all strains were identified and drug susceptibility was tested.The fragment including RRDR in rpoB gene of 46 multi-drug resistant strains and 15 pan-susceptible strains were analyzed by DNA sequencing.No mutation was found in 15 pan-susceptible strains.Thirteen types of mutations(including 9 point mutations,one insertion and 3 combine mutations)in the hot-point mutant region in rpoB gene were observed in 42 multi-drug resistant strains,with a rate of mutation of 91.30%.The sum rate of point mutations in codon 531 and that of codon 526 reached 69.57%.The finding suggests that the most frequency point mutations in multi-drug resistant isolates from Fujian Province were codon 531 and codon 526.
