Title: Studies of Protective Effect of Tedenan on Ultrastructure of Detrusor Cell Following Partial Outlet Obstruction in Rabbit Bladder
Abstract: Purpose To observe the protective effect of Tedenan on ultrastructure of detrusor cell in treating bladder dysfunction following outlet obstruction. Methods Twenty New Zealand white male rabbits were divided into four groups of 5 rabbits each.Control group was received operation but not forming obstructive model;other groups were received operation forming obstructive model.Two weeks after operation,Tadenan group received Tadenan orally at 100 mg/(kg·d) for three weeks;peanut oil group received vehicle only(peanut oil) and the obstruction group did not receive any drugs for three weeks.Five weeks after operation,each rabbit was anesthetized and the bladder was rapidly removed and weighted.The ultrastructure including rough endoplactmic reticulum and mitochondrion of detrusor cell of each bladder were examined in electron microscope. Results (1) Tadenan did not reduce the bladder mass but had protective effect on the capacity of bladder following partial outlet obstruction.(2) The average area of rough endoplactmic reticulum in Tadenan group was (1.259±0.274)μm 2 in each unit cytoplasm area(114.68 μm 2) versus (0.214±0.101)μm 2 in control group,(4.750±2.152)μm 2 in obstructive group,and (5.761± 1.775 )μm 2 in peanut oil group.(3) The relative density of mitochondrion was 1.292±0.164 in Tedenan group versus 1.582±0.286 in control group,0.973±0.073 in obstructive group and 0.925±0.102 in peanut oil group. Conclusions Tedenan have the protective effect on ultrastructure of detrusor cell.It is morphologic foundation of its protective effect on bladder function following partial outlet obstruction.
