Title: EFFECT OF PICROSIDEII ON INFLAMMATION REACTION AFTER CEREBRAL ISCHEMIA REPERFUSION INJURY IN RATS
Abstract: Objective To investigate the inhibitory effect of picrosideⅡon inflammatory reaction after cerebral ischemic reperfusion injury in rats.Methods Rat models of middle cerebral artery occlusion reperfusion were created by intraluminal thread method and divided into three groups in random.The rats in the treatment group received an injection of picrosideⅡ(10 mg/kg) via tail vein 2 hours after the ischemic reperfusion,while those in the sham-operation group and model group received sameamount of 0.1 mol/L PBS instead.The neurological behavioral function was evaluated by BEDERSON's test and the volume of cerebral infarction measured after tetrazolium chloride(TTC) staining.The apoptotic cells were counted by TUNEL assay and the expression of matrix metalloproteinase-9(MMP-9) in brain tissue was determined by immunofluoresce and Western Blotting assay.Results Abnormal neurological behavioral function was not found in rats of sham operation group with few apoptotic cells and weak expression of MMP-9.Significant neurological functional disorder was noticed in rats of the model group,together with cerebral infarction appeared in ischemic cortex and striatum,the amount of apoptosis increased,and the expression of MMP-9 enhanced.After picrosideⅡ treatment,the neuronal apoptosis and expression of MMP-9 significantly reduced,the neurological function improved and infarction volume decreased when compared with those in the control group.Conclusion PicrosideⅡ could inhibit the inflammation reaction against cerebral ischemia and reperfusion injury in rats.
