Title: Determination of the β-glucan Content in the Hullessbarley Straw Stalk
Abstract: Objective To establish a method to determine the β-glucan content in the Hullessbarley Straw stalk.Method Spectropho-tometry was used,Congo red was colorant.The optimal determination conditions were as follows:the acidity of solution pH was 8.0,reac-tion time was at 30 min,reaction temperature at 20 ℃ and the maximum absorption wavelength at 550 nm.Results The β-glucan had a good linearity in the range of 0~10 μg/mL(r =0.999 6) and the average recovery was 99.31 % with RSD of 1.29%.Conclusion The method was simple、accurate and reproducible.It is suitable for the determination of the β-glucan content in the Hullessbarley Strawstalk and other plants.
