Title: Determination of Polysaccharide in Pleurotus.citrinopileatus by Phenol-sulfuric Acid Method
Abstract: [Objective] The aim was to establish the determination method of polysaccharide in P.citrinopileatus.[Method] Determination of polysaccharide in P.citrinopileatus by phenol-sulfuric acid method was studied by two ways of single-factor experiment and orthogonal test.The main factor influencing the determination result was the amount of phenol,the amount of sulfuric acid was next,the water bath temperature was third and the water bath time was the smallest.The optimum experimental conditions were as follow: 0.2 ml phenol,5.0 ml sulfuric acid and 100 ℃water bath for 30 min.In the optimum experimental conditions,the standard solutions of glucose possessed good linear with absorbance when its concentration was 20-140 μg/ml.The linear equation was y=0.004 86x+0.094 6,and the correlation coefficient was 0.999 6.The color apparent system was special stability within 20-120 min.3.8% of mass fraction of polysaccharide extracted from P.citrinopileatus was obtained using the color apparent system and RSD=1.14%(n=6).[Conclusion] The study can provide reference for determination of polysaccharide content from P.citrinopileatus.
