Title: Determination of Total Polyphenols in Chestnut Shell by Folin-Ciocalteu Method
Abstract: [Objective]To explore a simple,scientific and accurate polyphenol analysis method. [Method]The optimum condition on determining total polyphenols content in chestnut shell by Folin-Ciocalteu method was studied. [Result] The results indicated that the optimum conditions of total polyphenols determination by Folin-Ciocalteu was as follow: 3. 0 ml of Folin-phenol reagent wae added in 0. 5 ml of sample and then6. 0 ml of 15% sodium carbonate was added. After the coloration reaction was 70 min at 40 ℃,optical density was measured at 760 nm. Undder the optimum conditions,the absorbanced linearly with the optical concentration of gallic acid in the range of 0 to 9 μg / ml( y = 0. 135 1x +0. 019 0,R2= 0. 999 3). The average spiked recovery rate was 99. 5%( RSD = 1. 2%). [Conclusion]This method is highly precise and stable,and suitable for determining total polyphenol content in chestnut shell.
