Title: Effect of high glucose on proliferation and injury of human peritoneal mesothelial cells
Abstract: Objective To investigate the effect of high glucose on the cell proliferation and the injury of human peritoneal mesothelial cells(HPMC).Methods The cultured HPMC were exposed to culture medium containing different concentrations of glucose(1.5%,2.5%,4.25%)for 6,12,24,48 hours.HPMC proliferation and the concentration of lactate dehydrogenase(LDH) in supernatants were detected by MTT assay and automatic biochemical analyzer respectively.Results(1)1.5%,2.5%,4.25% glucose inhibited the prolifernation of HPMC(P0.05).(2)1.5%,2.5%,4.25% glucose increased LDH expression in HPMC(P0.05),and the effect was time-and concentration-dependent.Conclusion High glucose can inhibit the proliferation and increase LDH expression of HPMC,and thus in part accelerate the progression of peritoneal injury and fibrosis.
