Title: Influence of caulis aristolochiae manshuriensis decoction on renal function and interstitial structure in rats at statutory dose according to pharmacopoeia
Abstract: Objective To investigate the influence of caulis tristolochiae manshuriensis (CAM) decoction on renal function and interstitial structure in rats at statutory dose according to Chinese pharmacopoeia. Methods Wistar rats were given daily dose of 1g/kg (equivalent to 0. 1g/kg in human)CAM decoction for 2 months by gavage. The parameters of renal function and renal histology were examined.Distribution of extracellular matrix (fibronectin) was also examined by immunohistological method. In addition, large dose (60g/kg) of CAM decoction was given to Wistar ras for 4 days, and serum creatinine and renal histology were also exandned. Results Large dose of CAM decoction induced acute renal failure in Wistar rats, tubular damage was the underlying lesion. However, in rats given with CAM decoction at dose according to pharmacopoeia, there were no statistical differences in serum creatinine, urinary activities of N-acetyl-beta-glucosaminidase (NAG ) and gamma-glutamyl transpeptidase (γ-GT ), urinary excretion of protein and glucose between CAM group and control group. Histological examination did not reveal significant abnormality in the kidney of CAM group. The architectare of renal interstitium was intact, and the distribution of fibronectin was just as same as control group. Condusion There is no significant adverse effect of CAM decoction on renal function and interstitial structure in rats at statutory dose according to pharmacopoeia.
