Title: A Study of the Association of MTHFR Gene Polymorphism with Cerebral Infarction
Abstract: Objective: To investigate the association of polymorphism of methylene tetrahydrofolate reductase(MTHFR) gene C677T and plasma total homocysteine(Hcy) with cerebral infarction in order to further confirm the effect of hereditary factors on the level of blood plasm Hcy and cerebral infarction and to provide scientific basis for the recognition of hereditarily susceptible genes in cerebral infarction,prevention and treatment of cerebral infarction.Methods: The level of blood plasm Hcy of the subjects was determined by highly efficient liquid chromatogra-phy and the genotype of C677T in MTHFR gene of 84 cases of cerebral infarction and 81 healthy controls was assayed by PCR-RLFP,with the frequency differences cmpared and analyzed in the distribution of genotype and alleles between the cerebral infarction group and the control group.Results: The level of blood plasma Hcy of those in the cerebral infarction group was significantly higher than that in the control group;the frequency of TT genotype and T alleles in the case group was remarkably higher than that in the control group,and the blood plasma Hcy level of TT genotype was notably higher than that of CT genotype and CC genotype in MTHFR C677T polymorphism.With all these differences having statistic significance(P 0.05).Conclusion: MTHFR C677T TT genotype raises the plasma Hcy level and it may be an important hereditary factor responsible for cerebral infarction.
