Bot. 50(7): 732–745. Illus. 1963.—This paper discusses the use of chemical reactions for specifically locating cellular constituents with the electron microscope. Presented in detail are results obtained by using alkaline hydroxylamine and ferric chloride as an electron stain for pectin. This treatment also results in the staining of certain cytoplasmic and nucleolar particles of unknown nature. Also described is the use of bismuth as an electron stain for nucleic acids. A series of micrographs is presented depicting bismuth stained onion root tip cells in various stages of mitosis.Read More
