Title: Modulation of IL-18 mRNA,TNF-α mRNA and IgE with Chinese herbal drugs in rats with bronchial asthma
Abstract: AIM:To explore the modulation of IL-18 mRNA,TNF-α mRNA and IgE with Chinese herbal drugs in rats with bronchial asthma. METHODS:Twenty-four Wistar rats were randomly divided into 3 groups:Control group,asthma model group and therapeutic group. Rat models of asthma were established using OVA and AI(OH)3. In atomization stimulating phase,a Chinese herbal solution of almond and 1iquoric root was used in therapeutic group,while normal saline was used in the other groups.Differential count in bronchoalveolar lavage fluid,quantification of IgE in serum and bronchoalveolar lavage fluids (BALF),and IL-18 mRNA and TNF-α mRNA levels in lung tissues were detected by immunoradiometric assay (IRMA) and RT-PCR. RESULTS:Compared with those in normal contro1,the serum IgE level,eosinophils percentage and expression of IL-18 mRNA and TNF-α mRNA significantly increased in model group. Compared with those in model group,significant reductions of serum IgE and expression of IL-18 mRNA and TNF-α mRNA were observed in therapeutic group. CONCLUSION:IL-18 and TNF-α may be involved in the occurrence of asthma. Chinese herbal solution inhibits the expression of IL-18 mRNA and TNF-α mRNA and thus alleviates the inflammation of asthma.
