To explore the physiological and pathological mechanisms of nuclear factor-κ-gene binding (NF-κB) changes induced by the mechanics effect of -Gx exposure by observing the NF-κB expression level in rabbit lung.
Methods
Twenty rabbits were randomly divided into control group and -Gx exposure group (experimental group) by random number table method. Each group had 10 rabbits. After exposing to the -Gx, the expression of NF-κB in rabbits′ lung tissue was detected by western blot and immunofluorescence. All data were processed by SPSS 22.0 statistical software package, and the measurement data were expressed by (±s). The difference on the positive cells in lung tissue slices was compared between control and experimental groups by t-test.
Results
By the fluorescence microscope observation, the positive cells expressed by NF-κB showed bright green fluorescence. Compared with the control group, the number of positive cells expressed by NF-κB in the lung biopsy of the experimental group was significantly increased (t=2.69, P<0.01). Western blot test results showed that the expression levels of NF-κB in lung tissue of experimental group were significantly increased as compared with that of control group (P<0.01).
Conclusions
-Gx exposure results in the increment of NF-κB expression in rabbits′ lung tissue. The up regulation of inflammatory factors and increased oxidative stress under -Gx exposure may indicate certain injury to lung.
