Title: Effects and significance of IL-8, IL-17 and TNF-α in bronchoalveolar lavage fluid of COPD rats with PM2.5
Abstract: Objective
To study the levels of BALF interleukin-8 (IL-8), IL-17 and tumor necrosis factor-α (TNF-α) in the chronic obstruction pulmonary disease (COPD), and role of COPD in the pathogenesis.
Methods
48 male Wistar rats were randomly divided into 2 groups: A group and B group, 24 rats in each group by tracheal infusion of the way, A, B two groups of rats by intratracheal instillation of sterile saline and PM2.5 were exposed 1 times every day, for 3 consecutive days, the dead animal in the last 24 hours after exposure, analysis of bronchoalveolar lavage fluid in IL-8, IL-17, TNF-α content of alpha.
Results
①Compared with A group of tracheal drip saline, the concentration of IL-8 in BALF of bronchoalveolar lavage fluid in B group with PM2.5 was significantly increased, and the difference was statistically significant (P<0.05). ②With the increase of rat exposure dose, the IL-8 level in BALF also increased, which was positively correlated; ③IL-17 and TNF-α levels in BALF of bronchoalveolar lavage fluid in COPD group were also increased, higher than IL-17, TNF-α level of tracheal drip into normal saline group, the difference between the two groups were statistically significant (P<0.05).
Conclusions
IL-8, IL-17, TNF-α may be involved in COPD airway and systemic inflammation, But IL-8 may be involved in COPD acute exacerbation of the occurrence and development of important inflammatory factors, IL-8, IL-17 and TNF-α interact with the occurrence and development of COPD in PM2.5 intervention, it is of great clinical significance to evaluate the severity and prognosis of COPD.
