Title: Screening of HIV-1 p24 Binding Peptides from Phage Display Peptide Library
Abstract: A phage display peptide library was screened by targeting recombinant p24 antigen.After three rounds of panning,eleven phage clones were chosen to test the binding affinity by ELISA,and ten of them were positive.DNA sequencing showed that nine of the positive clones display a unique sequence FTTRANA which has high affinity with HIV-1 p24 antigen.To improve the condition of phage ELISA,phage number and blocking buffer have been optimized,phage number ranged from 8.13×1011 to 1.02×1014 pfu/well and 0.5%BSA was highly recommended.Therefore,in this study,peptides combined with p24 antigen were obtained which might be useful in p24 antigen assay.
