Title: A new synthesis of 3-deazathymidine and of a related phosphoramidite synthon
Abstract: 3-Deazathymine (6), a DNA base analog, was synthesized in a new way. The β-selective glycosylation to the deoxynucleoside derivative and the use of the uncommon triisopropylphenyl-sulfonyl (TIPS) protecting group (→7) lead to a high yield of 3-deazathymidine (8). The TIPS-nucleoside derivative 9 is readily converted into the phosphoramidite 10, a suitable derivative for automatic DNA synthesis.
