Title: Establishment of fingerprinting for melon hybrids by SSR markers
Abstract: Two melon hybrids (Dongfangmi No.1 and 01-31) were analyzed by SSR marker to establish the standard DNA fingerprinting for seed test. Among 63 pairs of primers, 10 pairs with steady polymorphism were selected and distributed in melon genome. 2 alleles to 4 alleles were detected by each pair primers. There were 27 alleles detected in total. The average number of alleles detected was 2.7 per pair primer. The band size ranged from 85 to 270 bp. Finally, 7 out of 10 pairs of SSR primers were selected, which established fingerprinting for two hybrids. According to the data above mentioned, the fingerprinting of the melon hybrids was established and counted firstly. Then the paper introduced the method to calculate probability for the fingerprinting of each hybrid, and finally the standard, exclusive fingerprinting of melon hybrids was determined to distinguish the truth and falseness of samples in seed test. The experiment showed that SSR technique was practical and effective to identify melon hybrids.
