Title: Genetic Diversity in Three Different Natural Populations of Macrobrachium nipponense Revealed by SSR Markers
Abstract: To evaluate the genetic diversity and genetic differentiation of the different natural populations of Macrobrachium nipponense,11 microsatellite locus were used to analyze the genetic diversity in three natural populations of Macrobrachium nipponense Dongting,Poyang and Longgan. The results showed that 11 microsatellite locus were highly polymorphic. A total of 90 alleles were detected, each loci had 8. 182 alleles. In these natural populations the average number of allele( A) ranged from 32 to 38,and the value of average polymorphism information content( PIC) was 0. 590 7 to 0. 643 0,the value of average expected heterozygosity Hewas 0. 657 7 to 0. 705 5,the value of average observed heterozygosity Hoin three populations ranged from 0. 514 1 to 0. 579 9. Shannon indexes were greater than 1. Fiswere positive, average inbreeding coefficient of populations was 19%. FstValues show that the whole populational differentiation degree was lower. Most variation occurred within populations( 94. 62) among population( 5.38). The results showed that the genetic diversity is richer in three natural populations of Macrobrachium nipponense, and Poyang is the most abundant in genetic diversity population and the lowest level of genetic differentiation.
