Title: Effect of sub-hypothermia on changes of AIF expression after focal cerebral ischemia and reperfusion
Abstract: Objective To observe the effect of local sub-hypothermia on expression of AIF and study the relation of AIF releasing from mitochondria and DNA damage after focal cerebral ischemia and reperfusion injury.Methods The rats were randomly divided into sham-operation group,normothermia ischemia/reperfusion group and hypothermia ischemia/reperfusion group.The focal cerebral ischemia/reperfusion models of rats were achieved by modified thread embolism.After ischemia for 2 hours and reperfusion at middle cerebral artery for 3,6,12.24,72 hours and 7 days,the rats were killed.Neural functional defect evaluation was conducted before death.Sub-hypothermia at focal side was performed 30 minutes after 4-hour ischemia.Pathological changes were observed with hematoxylin and eosin.The expression of AIF was detected with immunohistochemical method.The apoptosis was detected with TUNEL.Results ①Assessment of neural functional defect:It was significant lower in the sub-hypothermia ischemic group than that in the normal ischemic group(P0.05).②AIF expressions:The expressions of AIF increased gradually with the extension of reperfusion time.It reached the peak at 72 hour.The expression in the sub-hypothermia ischemic group was less obviously than that in normal ischemic group(P0.05).③The detection of apoptotic cells:the apoptotic cells increased gradually with the extension of reperfusion time.It reached the peak at 72 hour.The expression in the sub-hypothermia ischemic group was lower than that in normal ischemic group(P0.05).Conclusion Sub-hypothermia plays an role in the AIF path of neuron apoptosis after suffering ischemic damage.
