Title: Expression of IL-13 receptor in leukemia Dami cells and influence of IL-13
Abstract: [Objective] To investigate whether Interleukine-13 receptor alpha 1 (IL-13Rα1) is expressed in Dami cells and to observe IL-13 effects on the expression of IL-13Rα1. [Methods] After Dami cells were cultured in serum-free medium for 20 h, total RNA was extracted from these cells. The first cDNA strand was synthesized by reverse transcription. Amplifications were performed, and then the PCR products were detected by agar gel electrophoresis. Data are analyzed by statistical analysis. [Results] Dami cells express IL-13Rα1 mRNA. The expression of IL-13Rα1 mRNA in Dami cells was lower in IL-13 treatment group (4 h) as compared with the serum treatment group, the mixture treatment group (IL-13 100 μg/L+10% serum) and the blank control group (P 0.05). But the expression of IL-13Rα1 mRNA in IL-13 treatment group (12 h) was not significantly different as compared with the serum treatment group, the mixture treatment group (IL-13 100 μg/L+10% serum) and the blank control group (P 0.05). IL-4Rα mRNA was expressed in Dami cells. However, IL-13 did not effect on the expression of IL-4Rα mRNA. [Conclusion] IL-13Rα1 mRNA is expressed in Dami cells. The expression of IL-13Rα1 mRNA is down-regulated by IL-13.
